Type

GHRP-2 (Growth Hormone-Releasing Peptide-2), also known as pralmorelin, is a synthetic hexapeptide that potently stimulates pituitary release of growth hormone by binding to the ghrelin receptor (GHS-R1a). It was approved in Japan in 2004 by the PMDA as a diagnostic agent for growth hormone deficiency assessment and is the only GHRP to hold any national regulatory approval — it is not FDA-approved. In the research and biohacking communities it is used for body composition improvement, recovery, and anti-aging applications.

Overview of GHRP-2

GHRP-2 selectively binds to the growth hormone secretagogue receptor (GHS-R1a) in the pituitary gland and hypothalamus, triggering GH release via Ca2+ influx, protein kinase C, and cAMP-dependent pathways. It also binds CD36, activating cytoprotective PI-3K/AKT1 signaling that reduces oxidative stress and inflammation independent of the GH axis.

growth hormone enhancement

  • Used as a diagnostic agent in Japan for assessing growth hormone deficiency in clinical settings.
  • Stimulates endogenous growth hormone release through ghrelin receptor activation.
  • Indirectly increases IGF-1 levels through enhanced GH secretion.

body composition

  • Enhanced muscle protein synthesis through elevated growth hormone and IGF-1 levels.
  • Improved lipolysis and body composition through GH-mediated fat oxidation.
  • Faster recovery from exercise and improved sleep quality.

protective effects

  • Myoprotective effects in muscle atrophy models through GHS-R1a stimulation.
  • Cytoprotective effects observed in cardiac tissue research.

Key benefits

Produces strong, pulsatile GH release from the pituitary by directly activating GHS-R1a, increasing peak GH levels more robustly than many other secretagogues

Promotes lean muscle mass accrual by elevating IGF-1 downstream of GH, supporting nitrogen retention and protein synthesis

Enhances lipolysis by shifting substrate utilization toward stored fat, improving body composition over extended cycles

Accelerates recovery from training and soft tissue injuries through GH-mediated upregulation of collagen synthesis and growth factors

Community interest

This peptide is still gaining traction in the community.

GHRP-2 Protocols

Subcutaneous injection is the primary and most effective route for GHRP-2, delivering precise GH-stimulating pulses with high bioavailability. Injections should be administered on an empty stomach — at least 2 hours post-meal — to prevent insulin from blunting GH release.

Reconstitution Instructions

Materials needed:

GHRP-2 lyophilized powder vial (5 mg or 10 mg)Bacteriostatic water for injection (2.5 mL per 5 mg vial)Insulin syringes (0.5–1 mL, 29–31 gauge)Alcohol swabsSharps disposal container

Steps to reconstitute

  1. Draw 2.5 mL of bacteriostatic water into an insulin syringe
  2. Insert the needle through the rubber stopper of the GHRP-2 vial at an angle and inject the BAC water slowly down the inner glass wall — never directly onto the lyophilized powder
  3. Do not shake the vial; gently swirl until the powder dissolves completely
  4. Cap the vial and store in the refrigerator at 2–8°C; use within 28 days
  5. Draw the desired dose volume into a fresh insulin syringe (e.g. 0.05 mL = 100 mcg at 2 mg/mL)
  6. Pinch a fold of skin at the injection site (abdomen, outer thigh, or upper arm), insert the needle at a 45–90 degree angle, and inject slowly
  7. Do not aspirate for subcutaneous injections; withdraw the needle at the same angle and apply gentle pressure
  8. Rotate injection sites at least 1 inch apart with each dose to prevent lipodystrophy

GHRP-2 Cycle

The GHRP-2 Cycle section explains how long a typical cycle lasts and what to expect during each phase. Over time, your body can become less responsive with continuous use.

Taking breaks between cycles may help maintain effectiveness and support better overall results.

Frequently asked questions

What is a peptide dosage calculator?

A peptide dosage calculator is a free tool that converts your vial size, bacteriostatic water volume, and target dose into an exact syringe draw volume. Instead of doing the reconstitution math by hand, you enter three inputs and instantly get the concentration of your solution and how many milliliters or syringeunits to draw. This calculator works for single peptide compounds and multi-peptide blends.

How do I calculate peptide dosage from a vial?

To calculate your peptide dose, divide the total peptide content of your vial in micrograms by the volume of bacteriostatic water you added in milliliters. This gives you your solution concentration in mcg/mL. Then divide your target dose by that concentration to get your draw volume. For example, a 5mg (5,000 mcg) vial reconstituted with 2mL of BAC water gives a concentration of 2,500 mcg/mL. A 250 mcg dose would require drawing 0.1mL. This calculator automates all of those steps instantly.

How much Bacteriostatic water should I add to a peptide vial?

Most people add 2mL to 3mL of bacteriostatic water per vial, but the right amount depends on the dose you want to draw and the syringe size you are using. Adding 1mL to a 5mg vial gives you a concentration of 5,000 mcg/mL, making each dose very small in volume. Adding 2mL gives you 2,500 mcg/mL, which is easier to measure on a standard insulin syringe. A general guideline is to choose a volume that puts your typical dose somewhere between 10 and 30 units on a U-100 syringe. Use the calculator above to test different water volumes and find what works for your dose.

How are peptides different from proteins?

Both are made of amino acids, but peptides are much smaller than proteins. Because of their tiny size, peptides can act like tiny messengers in the body, sending specific signals to your cells to tell them exactly what to do.
